The Valdosta-Lowndes County area was home to the Timucua, the Hitchitee, the Seminoles, and Creeks. By 1838 Native Americans were forced out of the south by the US government and pioneering settlers.

Lowndes County was officially created on December 23, 1825. In 1828 Franklinville was established as the county seat until it was moved to Troupville in 1837. The City of Valdosta, named after the plantation home of Govenor George Troup, was incorporated as the county seat in 1860.
